Branched PEG Derivatives for PEGylation

Branched PEG derivatives, also known as "Y-shaped" PEG derivatives, contain two linear methoxy PEG chain attached to a central core. The sterically bulky structure of these "Y-shaped" PEG derivatives will facilitate the single point attachment of the modified molecules.
The typical structure of "Y-shaped" branched PEG is as the following:

Branched PEG Derivatives for PEGylation

Maya now provides the following three kinds of "Y-shaped" PEG derivatives. Y-NHS-40K is used for amine PEGylation, Y-MAL-40K is used for thiol PEGylation, and Y-ALD-40K is for N-terminal PEGylation.
"Y-shaped" PEG derivatives are chemically purified during synthesis so that they are low in polydispersity and are essentially free of activated bifunctional PEG side products.

For Amine PEGylation


Y-NHS-40K: Y-Shape PEG NHS Ester, MW 40000

This "Y-shape" NHS ester will react with the amino group of lysine(s) or the N-terminal amine in biological active molecules to produce a stable amide linkage(s). This NHS ester will couple with the targeted molecules at pH 7-8. The PEGylation reaction can be finished in about 1 hour.
Average molecular weight of this product is 40,000 Da. Substitution of this product is 95% or greater. This product is basically free of activated bifunctional PEG side products.

Y-MAL-40K: Y-Shape PEG Maleimide, MW 40000

This "Y-shape" maleimide will react with the thiol groups in biological active molecules to generates a stable 3-thiosuccinimidyl ether linkage. This maleimide will couple with the targeted molecules at pH 5.0-6.5 in the presence of other functional groups.
Average molecular weight of this product is 40,000 Da. Substitution of this product is 90% or greater. This product is basically free of activated bifunctional PEG side products.

Y-ALD-40K: Y-Shape PEG Aldehyde, MW 40000

This "Y-shape" aldehyde will preferably react with the N-terminal amine in biological active molecules to produce a stable amine linkage in the presence of a reducing reagent such as sodium cyanoborohydride. This aldehyde will couple with the N-terminal amine of the targeted molecules at pH 5-8.
Average molecular weight of this product is 40,000 Da. Substitution of this product is 90% or greater. This product is basically free of activated bifunctional PEG side products.

Branched PEG derivatives, also known as "Y-shaped" PEG derivatives, contain two linear methoxy PEG chain attached to a central core. The sterically bulky structure of these "Y-shaped" PEG derivatives will facilitate the single point attachment of the modified molecules. Linear Monofunctional PEG Derivatives For PEGylation

Linear monofunctional PEG derivatives have a reactive group at one end of the PEG and the other end is capped with a methoxy group or a sugar group.

Linear monofunctional PEG derivatives have the following general structure:

where X is the functional group or active group and CAP can be either a methoxy group or a sugar molecule

Two patented high molecular weight linear monofunctional PEG derivatives, GLUC-NHS-35K and GALA-NHS-35Kare provided. They are both used for amine PEGylation.

For Amine PEGylation


The following linear PEG NHS esters as regular products meet with your different PEGylation requirements. Other PEG NHS esters with different molecular weights are available through custom synthesis.

GLUC-NHS-35K: Glucose PEG NHS Ester, MW 35000


This N-hydroxysuccinimide (or NHS) ester will react with the amino group of lysine(s) or the N-terminal amine in biological active molecules to produce a stable amide linkage(s). The presence of glucose group will increase the selectivity of the PEGylation reaction. GLUC-NHS-35K will couple to the targeted molecules at pH 7-8. The PEGylation reaction can be finished in about 1 hour.

The average molecular weight of GLUC-NHS-35K is 35,000 Da. Substitution is 90% or greater. This product is basically free of active difunctional PEG impurities.

M-PEG-MAL: Methoxy PEG Maleimide, MW 2000, MW 5000, MW 20000, MW 30000, MW 40000

M-PEG-MAL reacts with the thiol groups on cystein side chains in proteins and peptides to generates a stable 3-thiosuccinimidyl ether linkage. This maleimide will couple with the targeted molecules at about pH 5.0-6.5 in the presence of other functional groups. Substitution of M-PEG-MAL is 90% or greater.

GALA-NHS-35K: Glucose PEG NHS Ester, MW 35000

M-PEG-VS: Methoxy PEG Vinylsulfone, MW 5000, MW 20000

Sulfhydryl additon to the vinylsulfone generates a stabel beta-thiosulfonyl linkage. Vinylsulfone is stable in aqueous solution for a long time at high pH. Substitution of M-PEG-VS is 90% or greater. Purity of M-VS-PEG is 95% or greater.

M-PEG-VS: Methoxy PEG Vinylsulfone, MW 5000, MW 20000

M-PEG-SH: Methoxy PEG Thiol, MW 5000, MW 20000

M-PEG-ALD preferably reacts with the N-terminal amine in biological active molecules to produce a stable amine linkage in the presence of a reducing reagent such as sodium cyanoborohydride. This aldehyde will couple with the N-terminal amine of the targeted molecules at pH 5-8. Substitution of M-PEG-ALD is 95% or greater. Purity of M-PEG-ALD is 95% or greater.
